Nesitnoćelijski karcinom pluća predstavlja najčešći tip tumora pluća i javlja se u oko 85% obolelih. Postoje tri glavna histološka tipa nesitnoćelijskog karcinoma pluća: adenokarcinom, skvamocelularni karcinom i krupnoćelijski karcinom pluća (large cell). Bez obzira na nove pristupe lečenju ovih karcinoma petogodišnje preživljavanje ovih pacijenata je samo 15%. Osnovna polazišna tačka u odlučivanju u vezi sa lečenjem ove bolesti jeste tip karcinoma i stadijum bolesti u kojem je dijagnostikovan malignitet. Na žalost, s obzirom da je jedna od karakteristika nesitnoćelijskog karcinoma pluća da može da se razvija i asimptomatski, da ne postoji pouzdan skrining za ovo oboljenje najčešće se otkriva u kasnom stadijumu bolesti. Zbog ove činjenice, terapija je često neefikasna, što uzrokuje izuzetno visoku smrtnost obolelih. Trenutni istraživački pravci u vezi sa nesitnoćelijskim tumorima pluća koncentrisani su na otkrivanje novih biomarkera sa visokom senzitivnošću i specifičnošću – kako bi se poboljšala mogućnost rane dijagnoze ove bolesti. Upotreba novih biomarkera mogla bi da posluži i za razvoj novih terapeutskih pristupa. Otkrivanje nukleinskih kiselina koje potiču iz tumorskih ćelija u perifernoj cirkulaciji otvorilo je vrata razvoju novih neinvazivnih tehnika za otkrivanje ranih molekulskih biomarkera koji mogu biti korišćeni za dijagnozu malignih tumora, pa i nesitnoćelijskog karcinoma pluća. Gen za GAS5 dugu nekodirajuću RNK nalazi se na 1q25 lokusu: dužine je oko 650 baza i organizovan je u 12 egzona i 11 introna. Introni kodiraju male nuklearne RNK koje učestvuju u metilaciji – u epigenetskoj regulaciji. Egzoni kodiraju nekoliko varijanti GAS5 mRNK, ali zbog prisustva stop kodona, njihov rezultat nisu proteini, te se njihov transkript odbacuje. Ekspresija GAS5 je smanjena u različitim tipovima tumora, uključujući nesitnoćelijski karcinom pluća. Povećana ekspresija GAS5 indukuje apoptozu(programirana ćelijska smrt), a u ljudskom organizmu je povezana sa usporavanjem rasta tumora.

patients with this disease. There are three most common histological subtypes of nonsmallcell lung cancer: adenocarcinoma, squamocellular carcinoma, and large cell lung carcinoma. Regardless of the approach to treatment, most commonly done by targeted chemotherapy and radiotherapy, the five-year survival of these patients is only 15%. The main starting point in the treatment of lung cancer patients is the diseases stage, but since lung cancer is commonly unsymptomatic, it is usually discovered in one of the late stages. Consequently, the therapy is often inefficient, resulting in high mortality from this disease. Current research is directed at discovering new biomarkers for non-small-cell lung cancer with increasing sensitivity and specificity, with the aim of increasing the possibility of an early diagnosis and targeted treatment of this disease. The discovery of nucleic acids originating from cancer cells measurable in peripheral blood of lung cancer patients has opened the door to the development of new non-invasive techniques for early detection of molecular biomarkers which could be used for lung cancer diagnosis. The gene for the long non-coding RNA GAS5 can be found on the 1q25 locus: it is around 650 bases long and organized into 12 exones and 11 intrones. Intrones code small nuclear RNAs which have a part in DNA metillation – therefore in epigenetic regulation. Exones code several variants of GAS5 mRNA, but due to the presence of the STOP codons, their results are not proteins, therefore their transcripts are rejected. GAS5 expression is reduced in various types of tumors, including non-small-cell lung cancer. Increased GAS5 expression induces apoptosis, which in humans results in reduction of tumor growth. Objective The objective of this study was to explore the differences in long non-coding RNA GAS5 expression among patients in different stages of non-small-cell lung cancer and healthy population, the differences in GAS5 expression among different subtypes of non-small-cell lung cancer, and GAS5 expression depending on the stage of the disease...
